We are seeking an experienced individual to work as a Laboratory Manager at the National Institutes of Health in Bethesda, MD.

This is a long-term contract position which offers:

TASKS/SERVICES. The contractor shall:

**Provides technical experience needed to assist in studying the basic principles of plant and animal life**

+ Plan and administer biological research programs

+ Study basic principles of plant and animal life, such as origin, relationship, development, anatomy, and function

+ Identify, classify, and study structure, behavior, ecology, physiology, nutrition, culture, and distribution of plant and animal species

+ Develop and maintain liaisons and effective working relations with groups and individuals, agencies, and the public to encourage cooperative management strategies or to develop information and interpret findings

+ Collect, analyze and interpret data for all experiments performed

+ Search, retrieve and analyze publicly available data within the cBIOPortal for Cancer Genomics, COSMIC, dbSNP, dbVar, ClinVar, ExAC, The Human Protein Atlas, En-sembl, NCBI databases, DepMap, CCLE, GDSC, DGIdb, UniProtKBSwiss-Prot, and other online databases related to cancer, genomics, and proteomics

+ Make detailed and comprehensive records daily in laboratory notebook, to document: experimental purpose, results and interpretation thereof, and conclusions Laboratory notebook must adhere to NIH policies and lab-specific policies and will be reviewed on a regular basis by the principal investigator

+ Participate in and present research updates at weekly laboratory meetings, and on-on-one meetings with the Principal Investigator, for group and individual discussion

+ Adhere to all institutional policies, safety standards, and completion of appropriate training courses.

+ Perform PubMed and PubMed Central searches and other online searches to identify, read and assimilate scientific and biomedical literature relevant to projects; maintain a personal database of literature in EndNote.

+ Annotate gene and/or protein datasets using Ingenuity Pathway Analysis, g:Profiler, and/or DAVID.

+ Perform hierarchical clustering using Partek-- Genomics Suite--.

+ Perform and/or assist in highly accurate analysis of large-scale datasets.

+ Scan Western blots and perform densitometry, data quantification, and normalization

+ Develop X-ray films after exposure to Western blots.

+ Perform Bradford assays, Western blotting, immunoprecipitation, and co-immunoprecipitation.

+ Perform sequence alignments and sequence format conversions using bioinformatics tools including Clustal Omega, BLAST, and EMBOSS Seqret.

+ Perform macrodissection of fresh-frozen and/or paraffin-embedded human tissues and subsequent protein and nucleic acid extraction, purification, and quantification.

+ Perform protein and nucleic acid isolation and purification from mammalian cell cultures.

+ Perform molecular-based and cell-based assays including migration assays, viability assays, proliferation assays, wound-healing assays, kinase assays, reporter assays, drug sen-sitivity assays, siRNA/shRNA depletion, and in vitro ubiquitination assays.

+ Perform transient and stable transfections of plasmid DNAs into mammalian cell cultures.

+ Use BMG Omega plate reader and Bio-Rad Chemidoc Imager.

+ Perform cell counts using hemocytometer and automated cell counters including the Countess Automated Cell Counter.

+ Perform mammalian cell culture using sterile techniques; freeze stocks of mammalian cells for long-term storage and thaw stocks of frozen mammalian cells to initiate cultures.

+ Perform large-scale bacterial culture, bacterial transformation, bacterial colony growth and selection on agar plates, plasmid DNA isolation, plasmid DNA purification, and plasmid DNA quantification, and gel electrophoresis.

+ Design and generate mammalian expression constructs by site directed mutagenesis and cloning/subcloning.

+ Perform in silico predictions of sequence-variant function, using tools that include Poly-Phen-2, SIFT, Mutation Assessor, PROVEAN, SNAP2, PANTHER-PSEP, and Mut-Pred2.

+ Perform Sanger sequencing, sequence analysis, variant calling, and variant annotation in accordance with Human Genome Variation Society (HGVS) nomenclature.

+ Design and perform polymerase chain reaction (PCR) based assays and perform gel electrophoresis of PCR products.

+ Perform all research and related tasks in a meticulous manner, with scientific integrity, and including appropriate experimental controls, and following pre-approved experimental pro-tocol(s).

+ Perform tasks in a team-setting and/or individual setting, depending on project-specific needs.

**Assists with performing basic laboratory experiments and procedures**

+ Collect and analyze biological data about relationships among and between organisms and their environment

+ Supervise biological technicians and technologists and other scientists

+ Monitor and observe experiments, recording production and test data for evaluation by research personnel

+ Analyze experimental data and interpret results to write reports and summaries of findings

+ Perform all research and related tasks in a meticulous manner, with scientific integrity, and including appropriate experimental controls, and following pre-approved experi-mental protocols

+ Perform tasks in a team-setting andor individual setting, depending on project-specific needs

+ Ship, store, and dispose of biological and hazardous materials according to NIH guide-lines and policies

+ Assist with Western blotting

+ Assist with basic laboratory experiments and procedures

**Prepares, organizes and maintains various lab samples, supplies, and equipment**

+ Operate, calibrate, or maintain equipment used in quantitative or qualitative analysis, such as spectrophotometers, calorimeters, flame photometers, or computer-controlled analyzers

+ Maintain laboratory equipment

+ Operate laboratory equipment to analyze samples

+ Analyze laboratory findings

+ Perform Sanger sequencing, sequence analysis, variant calling, and variant annotation in accordance with Human Genome Variation Society HGVS nomenclature

+ Ensures lab complies with green lab program initiatives; may also help complete the green lab assessment if other point-of-contact not available.

+ Establish and maintain laboratory protocols, including SOPs.

+ Communicate in a timely, effective, and civil manner with NHGRI staff, service providers, and other internal and external personnel/stakeholders.

+ Monitor freezers daily for proper temperature; schedule service and/or repair of freezers that are out of normal temperature range; relocate contents of freezer to another location if freezer temperature is out of range
